Roof spots what can be done?

Cleaning my roof yesterday, Dirt and filth wash away easily with Dawn and soft bristle brush. Spots which cover 99% of roof membrane wouldn't come off or even lighten, even with scrubbing by hand.
Anyone have any knowledge of what caused the spots and what if anything I can or should do about the spots?

Could be tree sap or mold...Who cares? The harder you scrub the more of the wear layer you remove from the rubber fabric. Leave it alone like I do. My roof gets washed every time it rains...Nobody sees it or cares...Don

Exactly, I didn't want to rub a hole through the roof material so I didn't rub too hard. If its just sap stains I can live with that. Just wondering if there was anything I could or should do. Everything here in Georgia gets covered in a black mold when left outside, so I washed the dirt and mold off.

I usually give my roof a mild cleaning and treat with UV protectant. Same with the awning.
Living at 5000', there tends to be strong UV.
