Can I twist my glass sink bowl counter clockwise and remove it to find a leak behind it at the base?

I would suggest that if it leaks, remove the bowl completely.
But to answer your question, you may twist it but you probably wont find the leak.
It's best to start fresh.
I always liked the way the vessel sink looked, but was always a mess. I replaced ours with a new larger one that does not show the drips on the outside and is much easier to take care of.

You can not unscrew the vessel sink. To remove it you need to loosen and remove the drain under the sink. Then unscrew the nut on the tail piece, also under the sink, which is attached to the sink. The sink will then lift off, bring the tail piece with it.

The drain pipe has a lip on the top edge which is inside the glass bowl holding the bowl secure against the counter. As dbbls59 says, you must remove the drain pipe from under the counter and then you can lift up the sink from above.
